青新园蛛的生物生态学研究(英文)被引量:1

为调查青新园蛛的生物生态学特征,分别在田间和实验室条件下对其进行观察。如预期所示,幼蛛、亚成熟蛛和成蛛一起群居越冬。在湖南,青新园蛛每年可以繁殖3-4代,其中第4代寿命最长,第2-3代寿命最短。青新园蛛有织网的习性,雌蛛和雄蛛多次交配,一只雌蛛最多可产12个卵袋,每个卵袋平均含82.2颗卵粒。在群居模式中,刚孵化的蛛幼会安全的生活在雌蛛的背部。青新园蛛幼蛛的生长历期为40-120天,成蛛为33-176天。除第二代外,青新园蛛雌雄蛛的性别比率均小于1。该文详细描述了青新园蛛幼龄期和亚成熟期的生物学特征。在实验室条件下,青新园蛛的饥饿耐受力要强于抗旱力。

Biological Characteristics and Genome of Pardosa pseudoannulata

At the moment,little is found on the whole genomes of spiders in public databases.To this end,this study aimed to analyze the morphology,biological characteristics,and genome of Pardosa pseudoannulata,which was expected to add genome information of spiders.To be specific,Illumina sequencing was used to sequence the genome of this spider species,and K-mer(K=17)analysis to estimate the size,heterozygosity,and repetitiveness of the genome.More than 142.11 Gb high-quality reads were obtained,which were assembled into 3508.13 Mb contigs(N50=1035 bp)and 3542.56 Mb scaffolds(N50=1258 bp).Two peaks were found in the 17-mer distribution map.The genome of P.pseudoannulata was 4696.86 Mb,with the heterozygosity of 0.21%and 84.32%of repetitive sequences.This indicated that repetitive sequences were highly abundant in the genome of P.pseudoannulata and multi-generation selfing before genome sequencing can reduce the repetitiveness.In addition,NOVOheter can be used for genome assembly.This study layed a basis for the publication of spider genome data and provided a reference for the research on spider genomics.

Transcriptional response of Microcystis aeruginosa to the recruitment promoting-benthic bacteria被引量:1

Blooms of Microcystis aeruginosa occur frequently in many freshwater ecosystems around the world,but the mechanism of recovery has not been fully understood.In our previous study,three benthic bacterial species(E.sp013,Ba.spD06,and Ba.spD24)were identifi ed capable of promoting the recruitment of M.aeruginosa.Here,we further investigated the transcriptional response of M.aeruginosa to the benthic bacteria in early phase of recruitment by means of RNA-Seq analysis.In total,5803803 unigenes on average length of 404 bp were obtained from the transcriptome of M.aeruginosa.There were 54982 unigenes identifi ed as benthic bacteria-responsive unigenes based on the expression level analysis.Results of the protein-protein interaction analysis(PPI)show that the hub genes of the benthic bacteria responsive unigenes mediated network were ribosomal proteins of 30S and 50S,and the most signifi cant functional module of the network was related to the ribosome.Both the unigenes encoding the translation initiation factors(IF-2,IF-3)and elongation factors(lepA,fusA,and tufA)were up-regulated to respond benthic bacteria.Therefore,it indicates that the benthic bacteria have a positive infl uence on activating the ribosome during the early recovery stage of M.aeruginosa.

拟水狼蛛对镉和Bt蛋白联合作用的响应

用经镉(1.0mg/L)、Bt(Bacillusthuringiensis)蛋白(0.1mg/L)和Cd+Bt处理的果蝇饲喂拟水狼蛛(Pirata subpiraticus),测定拟水狼蛛体内Cd与Bt蛋白的富集状况和抗氧化酶表达水平;利用RNA-seq技术分析Cd+Bt处理拟水狼蛛的差异表达基因(DEGs),运用生物信息学方法构建DEGs的富集及调控通路。结果发现:与Cd和Bt处理的拟水狼蛛(Cd质量分数为(6.965±0.21)mg/kg、Bt质量分数为(3.82±0.28)μg/kg)相比,Cd+Bt处理的拟水狼蛛具有更高的Cd质量分数((12.23±0.86)mg/kg)和Bt质量分数((7.98±0.49)μg/kg)(P<0.05);与对照组、单一Cd和单一Bt处理的拟水狼蛛的相比,Cd+Bt处理还能提高拟水狼蛛体内的谷胱甘肽过氧化物酶、谷胱甘肽S-转移酶和超氧化物歧化酶的活性(P<0.05);通过对Cd+Bt处理和对照组进行转录组测序,得到6914条差异表达基因,并且与肽链内切酶活性、激素生物合成以及信息素代谢过程相关的基因(519条)都显著下调,共有14条参与卵母细胞减数分裂的基因在Cd+Bt处理下显著下调。转录因子和蛋白质互作分析表明,ZBTB等转录因子参与胁迫相关的调控网络,主要影响遗传信息的转录和翻译过程。
