The in vitro and in vivo anti-tumor effects of MTX-Fe_3O_4-PLLA-PEG-PLLA microspheres prepared by suspension-enhanced dispersion by supercritical CO_2被引量:1

The in vitro and in vivo anti-tumor efficacy of methotrexate-loaded Fe3O4-poly-L-lactide-poly(ethylene glycol)-poly-L-lactide magnetic composite microspheres(MTX-Fe3O4-PLLA-PEG-PLLA MCMs,MMCMs),which were produced by co-precipitation(C)and microencapsulation(M)in a supercritical process,was evaluated at various levels:cellular,molecular,and integrated.The results at the cellular level indicate that MMCMs(M)show a better anti-proliferation activity than raw MTX and could induce morphological changes of cells undergoing apoptosis.At the molecular level,MMCMs(M)lead to a significantly higher relative mRNA expression of bax/bcl-2 and caspase-3 than MMCMs(C)at 10μg mL-1(P<0.01);and the pro-caspase-3protein expression measured by Western blot analysis also demonstrates that MMCMs(M)can effectively activate pro-caspase-3.At the integrated level,mice bearing a sarcoma-180 tumor are used;in vivo anti-tumor activity tests reveal that MMCMs(M)with magnetic induction display a much higher tumor suppression rate and lower toxicity than raw MTX.Pharmacokinetic studies show that MMCMs(M)with magnetic induction significantly increase the accumulation of MTX in the tumor tissue compared with the other treatments.These results suggest that the MMCMs(M)prepared by the SpEDS process have great potential to play a positive role in the magnetic targeted therapy field.

采用超临界流体强制分散悬浮液技术,通过共沉淀法(C)和包埋法(M)成功制备了负载甲氨蝶呤(MTX)的Fe3O4-PLLA-PEG-PLLA磁性复合微球(MMCMs(C/M)),并从细胞、分子和整体水平对其体外及体内抑瘤活性进行评价.在细胞水平,MMCMs(M)比MTX具有更好的抗增殖活性,而且引起细胞形态学改变趋于凋亡.在分子水平,与10μg/mL MMCMs(C)相比,10μg/mL MMCMs(M)可导致Bax/Bcl-2和caspase-3的mRNA相对表达量显著增高(P<0.01);Western blot考察MG-63细胞的pro-caspase-3蛋白表达情况,结果显示,MMCMs(M)组可以有效地活化pro-caspase-3蛋白.在整体水平,构建S-180荷瘤小鼠作为动物模型,体内抑瘤实验结果表明,MMCMs(M)磁诱导组相对于MTX组具有较高的抑瘤活性和低毒性.药代动力学研究表明,MMCMs(M)磁诱导组MTX可以有效地积聚在肿瘤部位.综合实验结果表明,MMCMs(M)可在化学治疗药物磁靶向治疗领域发挥积极的作用.
